    Proteomics uncovers novel components of an interactive protein network supporting RNA export in trypanosomes

    In trypanosomatids, transcription is polycistronic and all mRNAs are processed by trans-splicing, with export mediated by noncanonical mechanisms. Although mRNA export is central to gene regulation and expression, few orthologs of proteins involved in mRNA export in higher eukaryotes are detectable in trypanosome genomes, necessitating direct identification of protein components. We previously described conserved mRNA export pathway components in Trypanosoma cruzi, including orthologs of Sub2, a component of the TREX complex, and eIF4AIII (previously Hel45), a core component of the exon junction complex (EJC). Here, we searched for protein interactors of both proteins using cryomilling and mass spectrometry. Significant overlap between TcSub2 and TceIF4AIII-interacting protein cohorts suggests that both proteins associate with similar machinery. We identified several interactions with conserved core components of the EJC and multiple additional complexes, together with proteins specific to trypanosomatids. Additional immunoisolations of kinetoplastid-specific proteins both validated and extended the superinteractome, which is capable of supporting RNA processing from splicing through to nuclear export and cytoplasmic events. We also suggest that only proteomics is powerful enough to uncover the high connectivity between multiple aspects of mRNA metabolism and to uncover kinetoplastid-specific components that create a unique amalgam to support trypanosome mRNA maturation

This seminar acknowledged the fact that in recent years, Digital Games (Computer-, Video- and Mobile-Games) have become an interesting topic for academic research. Within the booming sector of Game Studies the focus is no longer solely on the usage of digital games by children and teenager, or the influence digital games have on aggressive behaviour. Economic and cultural aspects of digital gaming play a significant role in the academic discourse on digital games.     Gender, race and parenthood impact academic productivity during the COVID-19 pandemic : from survey to action

    The coronavirus disease 2019 (COVID-19) pandemic is altering dynamics in academia, and people juggling remote work and domestic demands – including childcare – have felt impacts on their productivity. Female authors have faced a decrease in paper submission rates since the beginning of the pandemic period. The reasons for this decline in women’s productivity need to be further investigated. Here, we analyzed the influence of gender, parenthood and race on academic productivity during the pandemic period based on a survey answered by 3,345 Brazilian academics from various knowledge areas and research institutions. Productivity was assessed by the ability to submit papers as planned and to meet deadlines during the initial period of social isolation in Brazil. The findings revealed that male academics – especially those without children – are the least affected group, whereas Black women and mothers are the most impacted groups. These impacts are likely a consequence of the well-known unequal division of domestic labor between men and women, which has been exacerbated during the pandemic. Additionally, our results highlight that racism strongly persists in academia, especially against Black women. The pandemic will have long-term effects on the career progression of the most affected groups. The results presented here are crucial for the development of actions and policies that aim to avoid further deepening the gender gap in academia

    Plankton Community Respiration and ETS Activity Under Variable CO2 and Nutrient Fertilization During a Mesocosm Study in the Subtropical North Atlantic

    The enzymatic electron transport system (ETS) assay is frequently used as a proxy of respiratory activity in planktonic communities. It is thought to estimate the maximum overall activity of the enzymes associated with the respiratory ETS systems in both eukaryotic and prokaryotic organisms. Thus, in order to derive actual respiration rates (R) from ETS it is necessary to determine empirical R/ETS conversion algorithms. In this study we explore the temporal development of R and ETS activity in natural plankton communities (from bacteria to large phytoplankton) enclosed in mesocosms, treated with different CO2 concentrations. The experiment lasted 30 days, during which abrupt changes in community structure and biomass occurred through a sharp transition from oligotrophy (phase I) to highly eutrophic conditions (phase II) after nutrient-induced fertilization (day 18). R and ETS did not show any response to CO2 under oligotrophic conditions, but R increased significantly more in the two high CO2 mesocosms after fertilization, coinciding with a sharp rise in large phytoplankton (mostly diatoms). R and ETS were significantly correlated only during the eutrophic phase. The R/ETS ranged more than threefold in magnitude during the experiment, with phase-averaged values significantly higher under oligotrophic conditions (0.7-1.1) than after nutrient fertilization (0.5-0.7). We did not find any significant relationship between R/ETS and community structure or biomass, although R correlated significantly with total biomass after fertilization in the four mesocosms. Multiple stepwise regression models show that large phytoplankton explains most of the variance in R during phases I (86%) and II (53%) and of ETS (86%) during phase II, while picophytoplankton contributes up to 73% to explain the variance in the ETS model during phase I. Our results suggest that R/ETS may be too variable in the ocean as to apply constant values to different communities living under contrasting environmental conditions. Controlled experiments with natural communities, like the present one, would help to constrain the range of variability of the R/ETS ratio, and to understand the factors driving it

    Southern Ontologies. Reorienting Agendas in Social Ontology

    This article addresses ontological negotiations in the Global South through three case studies of community-based research in Brazil and Ghana. We argue that ontological perspectives of Indigenous and other subjugated communities require an ontological pluralism that recognizes the plurality of both representational tools and ways of being in the world. Locating these two readings of ontological pluralism in the politics of the Global South, the article highlights a wider dynamic from ontological paternalism to ontological diversity to ontological decolonization. We conclude by arguing that this dynamic provides important lessons for reorienting agendas in social ontology through Southern Ontologies

    Impact of Musashi-1 and Musashi-2 Double Knockdown on Notch Signaling and the Pathogenesis of Endometriosis

    The stem cell marker and RNA-binding protein Musashi-1 is overexpressed in endometriosis. Musashi-1-siRNA knockdown in Ishikawa cells altered the expression of stem cell related genes, such as OCT-4. To investigate the role of both human Musashi homologues (MSI-1 and MSI-2) in the pathogenesis of endometriosis, immortalized endometriotic 12-Z cells and primary endometriotic stroma cells were treated with Musashi-1- and Musashi-2-siRNA. Subsequently, the impact on cell proliferation, cell apoptosis, cell necrosis, spheroid formation, stem cell phenotype and the Notch signaling pathway was studied in vitro. Using the ENDOMET Turku Endometriosis database, the gene expression of stem cell markers and Notch signaling pathway constituents were analyzed according to localization of the endometriosis lesions. The database analysis demonstrated that expression of Musashi and Notch pathway-related genes are dysregulated in patients with endometriosis. Musashi-1/2-double-knockdown increased apoptosis and necrosis and reduced stem cell gene expression, cell proliferation, and the formation of spheroids. Musashi silencing increased the expression of the anti-proliferation mediator p21. Our findings suggest the therapeutic potential of targeting the Musashi-Notch axis. We conclude that the Musashi genes have an impact on Notch signaling and the pathogenesis of endometriosis through the downregulation of proliferation, stemness characteristics and the upregulation of apoptosis, necrosis and of the cell cycle regulator p21

    Transdisciplinary Philosophy of Science: Meeting the Challenge of Indigenous Expertise

    Transdisciplinary research knits together knowledge from diverse epistemic communities in addressing social-environmental challenges, such as biodiversity loss, climate crises, food insecurity, and public health. This paper reflects on the roles of philosophy of science in transdisciplinary research while focusing on Indigenous and other subaltern forms of knowledge. We offer a critical assessment of demarcationist approaches in philosophy of science and outline a constructive alternative of transdisciplinary philosophy of science. While a demarcationist focus obscures the complex relations between epistemic communities, transdisciplinary philosophy of science provides resources for meeting epistemic and political challenges of collaborative knowledge production
